Offer Description
The work is part of the Knowledge Generation project (PID2023-146767OB-00). The objective is to identify the mechanisms responsible for the direct reprogramming of hGEO and to modify them, in order to achieve efficient reprogramming and transplantation viability to an animal model of spinal cord injury. The activities to be performed for the position include:
1. Expression of the apoptosis blocking gene BCL2 together with the proneural gene NeuroD1.
2. Suppression of the repressor of transcription (REST) complex in hGEO.
3. Lentivirus packaging.
4. REST gene deletion by CRISPR/Cas9 in hGEO.
5. Cotransfection of hGEO with lentivirus.
6. Cell culture.
7. Analysis of neuronal markers by immunofluorescence, RT-qPCR, and Western blot.
8. Analysis of neuronal activity by patch-clamp.
